The spider is Helen. It represents how he now views his partner and committed relationships with women--as a spider luring him in to a trap of responsibility and commitment. Basically there's a part of him that wants to continue to live his past life as a player (as represented by the actor version), and in trying to reconcile that with his current self (the responsible professor), he blames her for 'forcing' him in to something that part of him doesn't want (the actor version).
The entire movie is about one man's psyche and trying to reconcile all of these conflicting emotions/desires. Much of it isn't literal; it's a symbolic movie. When the actor personality crashes the car with the mistress, it represents him trying to 'kill' that part of his personality in an attempt to commit to Helen. This was the catalyst that caused him to see Helen as a spider. But as you see by the end, he hasn't entirely killed off the actor personality, and to the extent that he has, in his resentment he now views her as a spider that finally lured him to the center of the web, and thereby crushing the will of the actor personality.
The movie is from an entirely masculine perspective, and deals with male anxieties about 'settling down' and losing vitality. He's revolting against becoming the professor archetype and suffering psychological 'splitting', but he loses the 'battle' by the end. The key to the crush show represents devaluation (vs. idealization)--a preventative measure against deleterious narcissistic rage.
